The aim of this work was the development of predictive quantitative structure–activity relationships (QSARs) in order to predict the acute toxicity of pesticides in earthworms (Eisenia foetida). Experimental toxicity data were obtained from the Pesticide Properties Database (PPDB) developed at the University of Hertfordshire. Molecular structures were drawn with ACD ChemSketch. Non-conformational molecular descriptors were calculated using PaDEL, Mold2 , DataWarrior and QuBiLs-MAS programs. After excluding the constant, linearly dependent, and unique value descriptors, a total of 2,055 structural variables were obtained. The Balanced Subset Method (BSM) was used to divide the molecular set (58 molecules) into 3 groups for calibration, validation and prediction. QSAR linear regressions of 1-5 descriptors were carried out using a mathematical algorithm, the Replacement Method. A 4-descriptor linear regression model was found to have the best validation results. This model was then used to predict the toxicity of 21 pesticides whose experimental values were unverified or less reliable. The predictions were generally close to the unverified reported values.
