Viticulture and enology are undergoing rapid growth in Entre Ríos, and the use of indigenous yeasts is becoming more widespread to obtain distinctive wines. The main objective was to analyze two winemaking processes with regional grape varieties (Tannat and Marselan) using indigenous yeasts (Saccharomyces cerevisiae, GenBank OQ520880.1) previously isolated. One system was inoculated with free cells and the other with cells immobilized in an alginate matrix. The experiment consisted of a 22 experimental design. Process monitoring was performed by soluble solids determination, density by an empirical formula and the overall rate of substrate consumption for each system. Assays carried out on Tannat variety showed that immobilized cells exhibited a greater capacity to metabolize sugars (7.90 ± 0.19 g·L⁻¹·d⁻¹) compared with the same variety using free cells (6.59 ± 0.13 g·L⁻¹·d⁻¹). However, no statistically significant differences were observed between the free system (6.75 ± 0.16 g·L⁻¹·d⁻¹) and the immobilized cells (6.75 ± 0.11 g·L⁻¹·d⁻¹) when compared with Marselan variety. In conclusion, immobilization of indigenous yeasts could provide technological advantages to the winemaking process.
