This study aimed to contribute to the understanding of the ecology and distribution of ticks with potential epidemiological significance. Species richness, Shannon Diversity Index, prevalence, and abundance of ticks associated with Akodon montensis Thomas and Oligoryzomys nigripes Olfers, in Misiones, Argentina were analyzed. Samples from 726 rodents collected between 2007 and 2018 at 21 sampling sites, distributed across two phytogeographic regions and seven areas (I-VII) were examined. 42 ticks (Ixodida, Ixodidae), were identified: 22 larvae and 18 nymphs of Amblyomma ovale Koch from both rodents, one Amblyomma brasiliense Aragão nymph from A. montensis, and one Ixodes sp. nymph from O. nigripes. The specific diversity was 0.22, prevalence 4.13 %, and mean abundance 0.06. Significant differences were observed in tick prevalence and abundance between phytogeographic areas of the Planaltense sector and the Misionero sector. In area V (Misionero sector), A. ovale showed higher prevalence in O. nigripes (33 %). The first association of A. brasiliense with A. montensis in South America is recorded, as well as the association between Ixodes sp. and O. nigripes in Misiones.
