Resumen

Atriplex lampa is a valuable shrub from Patagonia. We isolated plant growth-promoting rhizobacteria (PGPR) from its rhizosphere in steppe soils (PS) and coastal dunes (Du) and assessed their effects on plant growth and rhizobacterial community. PGPR isolates produced ammonia, indoleacetic acid, and solubilized phosphorus. Du strains were affiliated with salt-tolerant Stutzerimonas and Halomonas, while PS strains included Bacilli and Actinomycetes. Stutzerimonas sp. 5A and Pseudarthrobacter sp. 15 inoculation significantly increased plant height and biomass, whereas Bacillus sp. 48 had no effects. The rhizobacterial community structures changed after inoculation, while their diversity remained unaffected. Nitrogen-fixing and growth-promoting families (Azospirillaceae, Beijerinckiaceae, and Pseudomonadaceae) increased in roots inoculated with 5A and 15. Near full-length 16S rRNA gene sequences of each isolate (5A, 15, and 48) were used to identify their corresponding OTUs within the V4 region amplicon dataset. The absence of significant differences in such OTU abundances among treatments indicated that inoculation triggered taxonomic shifts rather than promoting their own enrichment. Our results not only confirm the biotechnological potential of strains 5A and 15 but also provide the novel insight that PGPR can promote native dryland plant growth by reshaping the rhizosphere bacterial community structure, rather than solely through enhancing their own proliferation.

Métodos

The study was conducted in two Atriplex lampa (Moq.) D. Dietr. occurring sites by the coast of the Chubut Province, near Puerto Madryn City (Argentina). One of them was a site in the Patagonian steppe close to a cluster of fishing industries, which used to receive fish-processing effluent discharges. Vallejos et al. (2020) sampled and characterized soils under A. lampa patches during the dumping of effluents in this site (ES1). Four years after discharge cessation (2021), we sampled the soil under A. lampa patches of such site again (ES2). The mean annual temperature and the mean annual precipitation are 13.3 ◦C and 185.35 mm, respectively (1971–2021 time series, INTA SIPAS, http,//sipas.in ta.gob.ar/). Soils are a complex of Typic Torriorthents (Pereyra and Bouza, 2019). In addition, we sampled another A. lampa occurring area, the coastal dunes near Puerto Madryn (Du). The soil composition of coastal dunes is homogeneous and mostly includes fine sand. The main source of sediments is the erosion of cliffs surrounding the Nuevo gulf (Bunicontro et al., 2017). At each site, five A. lampa patches were randomly selected, and three upper soil sub-samples (0–10 cm depth and 10 cm in diameter) were collected. Soil samples were immediately transported to the laboratory at 4 ◦C. Sub-samples were pooled, homogenized, and sieved through a 2 mm mesh for further processing.
