Punta Lara Nature Reserve (RNPL) is located on the coast of the “Río de La Plata” estuary and biogeographically belongs to the Delta del Paraná district (Esteros del Iberá province). In a previous survey, 23 species of ferns and one lycophyte were recorded. Periodic inventory updates enable the comparison and interpretation of changes in the biological composition over time, which can be reflected in species disappearance or new records. The aim of this work was to provide a synopsis of the fern and lycophyte taxa inhabiting RNPL, more than ten years after the publication of the first inventory of these plant lineages in the Reserve, updating information on nomenclatural and systematic aspects, along with biogeographic patterns. We conducted periodic surveys in different habitats within the reserve, taking photographic records, habitat data, and estimating the species abundance. A key was developed based on the taxa found. There was an increase in the number of fern taxa recorded, reaching 28. The five species of ferns not cited in previous inventories are Amauropelta platensis, Asplenium clausenii, Cyclosorus interruptus, Cyrtomium falcatum, and Pleopeltis pleopeltifolia. We propose a new combination of Rumohra calahuala and confirm the establishment of Adiantum capillus-veneris in natural environments, granting it naturalized status. The RNPL’s fern and lycophyte flora has Parana-Atlantic components mixed with species mainly from the Chaco and Pampas regions, forming a spectrum of different communities. As a contribution to the management plan of the Reserve, studies on the reproductive biology of ferns and the cultivation of sporophytes are carried out to reinforce populations decreasing in number, if necessary.
