Resumen

This data resource provides information on the diversity and distribution of mammals in the province of Tierra del Fuego, Antarctica, and the South Atlantic Islands, using the technique of environmental DNA metabarcoding in freshwater environments. It includes a reference database of the mitochondrial 12S rRNA gene, constructed from tools such as GenBank and de novo sequences from local mammals. Additionally, it features sequencing data generated with the Illumina MiSeq system.

Métodos

A local database of 12S rRNA (12S) gene sequences was generated for mammal species present in Tierra del Fuego. Some of these sequences were obtained from the GenBank genetic database, while sequences for species with no available information were generated from tissue samples stored in the sample bank of the Molecular Ecology Laboratory at the Austral Centre for Scientific Research (CADIC-CONICET). DNA amplification was performed using the 12S primers XL: 5'-AAAGCGAGGCACTGAAAATG-3' and XH: 5'-GGTGTAAGCCGGATGCTTT-3' (Poljak, unpublished), resulting in a 770 base pair (bp) fragment of the 12S region. Five sampling sites were selected across the province: the Grande River, located in the Magellanic Steppe region; the Ewan River, within the Ecotone; and the Milna, Lasifashaj, and Olivia Rivers, situated in the Andean Cordillera. Finally, eDNA extraction was followed by high-throughput sequencing using the Illumina MiSeq platform.
