Antimutagenic properties of goat's probiotic against mutagens

Resumen

Gastrointestinal tract microbial ecology has an important effect on goat health and performance. Mutagens are frequently formed during stress or bacterial gastrointestinal infections and by endogenous microflora. The ability of goat´s probiotic to reduce fecal mutagens and cancer marker was previously determined. Binding of mutagens to microbial cells has been suggested to be a possible mechanism of antimutagenicity. The ability of the goat´s probiotic Lactobacillus reuteri DDL 19, L. alimentarius DDL 48 and E. faecium DDE 39, B. bifidum DDBA, and the mixture of goat probiotics (MGP) was constituted by the mentioned strains in a relation 1:1:1:1 was investigated at different cells concentration (1-2.106, 1-2.108, and 1-2.1011 CFU ml-1). The antimutagenic activity of these strains against benzopirene B[a]P and sodium azide (SA) were determined, using S. typhimurium TA 100. In presence of sodium azide the number of revertants of S. typhimurium TA 100 were 503±9. The natural revertants in the media whitout mutagens were 103±6. In presence of different probiotic concentrations (1.106 1.10 8, 1.1011 CFU/mL ), the number of revertants diminished: 427±1, 361±1, 215±8 to L. reuteri DDL 19; 454±6, 396±8, 251±9 to L. alimentariusDDL48; 350±11, 313±4, 391±2 to B. bifidum DDBA; 396±6, 380±7, 242±4 to E. faecium DDE39; and 363±2, 295±7, 193±6 to MGP. In presence of benzopirene the Number of cells revertants of S. typhimurium TA 100 were 328±9. The natural revertants in the media whitout mutagens were 160±8. In presence of different probiotic concentrations (1.106 1.10 8, 1.1011 CFU/ml ), the number of revertants diminished: 297±9, 280±5, 209±9 to L. reuteri DDL 19; 302±6, 294±6, 259±9 to L. alimentariusDDL48; 300±7, 275±8, 209±10 to B. bifidum DDBA; 288±4, 268±5, 221±9 to E. faecium DDE39 and 281±8, 245±6, 200±3 to MGP. The number of revertants of S. typhimurium TA 100 decreases significantly due the increment to the cells number. However, B. bifidum DDBA, reach the maximal desmutagenicity to SA at 1.108 CFU ml-1. In conclusion the amount of SA and B[a]P bound was strain, mutagens and cells concentration specific. The potential antimutagenic properties of MGP demonstrated after this consumption by goats, could be explained by the binding of mutagens.
